چکیده

The ectomycorrhizas (ECM) formed by Lactarius quietus, an ECM fungus specifically associated with Quercus spp., are abundant all the year long. Root and stem growth, beginning before bud break in oak, are strong carbon sinks partially fulfilled with carbohydrate reserves. We hypothesized that L. quietus contributes to providing trees with carbon at bud break through enzymatic activities before photosynthesis begins. Activities of eight secreted enzymes (xylosidase, glucuronidase, cellobiohydrolase, b-glucosidase, N-acetyl-glucosamine, leucine aminopeptidase, acid phosphatase and laccase) relevant to carbon cycling and the release of phophorus and nitrogen from soil organic matter were measured on L. quietus ECMs before, during and after the bud break. Phenological, climatic and pedoclimatic parameters were also measured. Laccase, glucuronidase, cellobiohydrolase and b-glucosidase activities proved to be significantly related to tree reactivation and climate. All these activities can help the formation of new tissues by supplying carbon. L. quietus can behave saprotrophically, using soil organic matter as substrate. This is consistent with the hypothesis that it provides the oak trees with carbon when demand is high and photoassimilates are not yet available. r 2007 Elsevier Ltd. All rights reserved.

Gene transcription in Lactarius quietus-Quercus petraea ectomycorrhizas from a forest soil.

Extracting fungal mRNA from ectomycorrhizas (ECMs) and forest soil samples for monitoring in situ metabolic activities is a significant challenge when studying the role of ECMs in biogeochemical cycles. A robust, simple, rapid, and effective method was developed for extracting RNA from rhizospheric soil and ECMs by adapting previous grinding and lysis methods. The quality and yield of the extra...

Study of Phenological Changes in Black Locust using Descriptive Methods and Quantitative Analysis

Studying the phenological stages and determining their occurrence in different biological conditions is useful instrumental in restoring and selecting the bases in forestry. Therefore, descriptive method and quantitative analysis were used to study the phenology of Black Locust (Robinia pesudoacacia L.). In descriptive method, the phenomenological observations were recorded during the speci...

Environmental drivers of ectomycorrhizal communities in Europe's temperate oak forests.

Ectomycorrhizal fungi are major ecological players in temperate forests, but they are rarely used in measures of forest condition because large-scale, high-resolution, standardized and replicated belowground data are scarce. We carried out an analysis of ectomycorrhizas at 22 intensively monitored long-term oak plots, across nine European countries, covering complex natural and anthropogenic en...

Frost tolerance and bud dormancy of container-grown yellow birch, red oak and sugar maple seedlings.

Container-grown seedlings of red oak (Quercus rubra L.), sugar maple (Acer saccharum Marsh.) and yellow birch (Betula alleghaniensis Britton) in their first year of growth were overwintered outdoors. Tolerance of roots and stems to freezing was compared from late summer to the following spring. Mitotic activity in the apical bud was related more closely to air temperature than to bud dormancy a...

Changes of free polyamines in the leaves and stems of ‘Kinnow’ mandarin tree as affected by alternate bearing

In order to investigate the seasonal changes and the possible role of the free polyamines in the leaves and stems on the alternate bearing habit of the ‘Kinnow’ mandarin (Citrus reticulata Blanco) trees, a comparative study was conducted to analyze the levels of free polyamines  (putrescine, spermidine and spermine) in the leaves and stems tissues of “on” and &ldquo...
